RSM-Based Optimization of Fermentation Conditions and Kinetic Studies of Glutamic Acid and Lysine Production by Corynebacterium glutamicum

Corynebacterium glutamicum is an authenticated microorganism that supports amino acid production consistent with dietary importance. Fermentation parameters like temperature, agitation speed, and carbon source concentration were optimized using response surface methodology. Surface model suggested optimal fermentation including 30°C, 50 g/L glucose concentration, shaking speed of 120 rpm furnished 14.2 glutamic 5.1 lysine, comparable the predicted values. After optimizing in shake flasks, kinetics was studied a stirred fermenter. The kinetic study revealed substrate consumption rate achieved maximum level 3.36 g/L/hour between 12 18 hours; afterwards, it decreased fell to 1.9 g/L/hour. Average biomass yield over period 48 hours 0.337 g/L; however, 0.51 noted 10 20 hours. Overall molar coefficient CO2 ( Y C CO 2 / S ) found be 0.234. id="M2"> X 0.6 exponential phase which afterwards. This indicates average time g/L, within obtained under conditions. id="M3"> id="M4"> near zero at hrs.
